Characterization of an ultraviolet B-induced lipase in Arabidopsis.

چکیده
An Arabidopsis expressed sequence tag clone, 221D24, encoding a lipase has been characterized using an antisense approach. The lipase gene is expressed during normal growth and development of Arabidopsis rosette leaves but is down-regulated as the leaves senesce. When plants are exposed to sublethal levels of UV-B radiation, expression of the lipase is strongly up-regulated. The lipase protein is localized in the cell cytosol and is present in all organs of Arabidopsis plants. Recombinant lipase protein produced in Escherichia coli preferentially hydrolyzed phospholipids, indicating that the gene encodes a phospholipase. Transgenic plants in which lipase expression is suppressed showed enhanced tolerance to UV-B stress but not osmotic stress and were unable to up-regulate PR-1 expression when irradiated with UV-B. The observations collectively indicate that the lipase is capable of deesterifying membrane phospholipids and is up-regulated in response to UV-B irradiation.

Molecular Cloning and Characterization of a Lipase from an Indigenous Bacillus pumilus
Cloning and sequencing of a lipase gene from an indigenous Bacillus pumilus, strain F3, revealed an open-reading frame of 648 nucleotides predicted to encode a protein of 215 residues. Sequence analysis showed that F3 lipase contained a signal peptide composed of 34 amino acids with an H domain of 18 residues. A tat-like motif was found in the signal peptide similar to some other Bacillus pumil...

متن کاملIdentification and Characterization of LHCB1 Co-Suppressed Line in Arabidopsis
To explore the function of light-harvesting complex protein (LHCP) in Arabidopsis growth and development, the Leclere and Bartel seed collection was screened. In this collection randomly cloned cDNAs are expressed under the CaMV35S promoter. A pale green line has been identified and characterized in more details. Analysis of the inserted cDNA in the pale green line showed it encodes LHCB1 prote...
متن کاملCharacterization of Arabidopsis photolyase enzymes and analysis of their role in protection from ultraviolet-B radiation.
DNA photolyases are enzymes which mediate the light-dependent repair (photoreactivation) of UV-induced damage products in DNA by direct reversal of base damage rather than via excision repair pathways. Arabidopsis thaliana contains two photolyases specific for photoreactivation of either cyclobutane pyrimidine dimers (CPDs) or pyrimidine (6-4)pyrimidones (6-4PPs), the two major UV-B-induced pho...
